    利用RecTE基因重组体系构建hrpS基因缺失型冠菌素工程菌株

    Application of RecTE recombination system to construct hrpS gene deletion coronatine engineering strain

    • 摘要: 冠菌素 (coronatine, COR) 是一种新型植物生长调节剂,能有效调控植物生长,增强植物抗逆性。目前冠菌素主要通过微生物菌株发酵的方式获得,而菌株自身产率低是限制冠菌素应用的最大问题。为了开发冠菌素高产菌株,本研究以模式菌株丁香假单胞菌番茄致病变种Pseudomonas syingae pv. tomato DC3000为出发菌株,利用RecTE基因重组体系对菌株Pst DC3000中hrpS基因进行了敲除,并成功筛选到基因重组菌株H1-20。对H1-20菌株进行发酵培养,结果表明,H1-20菌株中冠菌素的产量达到了22.67 mg/L,是出发菌株冠菌素产量的2.36倍。本研究通过RecTE基因重组体系,成功地构建了冠菌素工程菌株H1-20,为产冠菌素菌株提供了更多选择,为后续进一步开展冠菌素高产菌株改造工作奠定了基础。

       

      Abstract: Coronatine (COR) is a new plant growth regulator, which can effectively regulate plant growth and enhance plant stress resistance. At present, coronatine is mainly obtained by fermentation of microbial strains, but the low yield of strains is the main problem limiting the application of coronatine. To develop high-yield strains of coronatine, the RecTE genetic recombination system was used to knockout hrpS gene in model strain Pseudomonas syingae pv. tomato DC3000, and the mutant strain H1-20 was selected from transformants. After fermentation culture for H1-20, the result showed that the coronatine production of H1-20 reached 22.67 mg/L, which was 2.36 times to Pst DC3000. In this study, engineering strains were successfully constructed by the application of RecTE recombination system, which provided more options for coronatine producing strains and laid a foundation for further development of high-yielding strains of coronatine.
