    脱镁叶绿酸钠对胡萝卜灰霉病菌的抑制作用

    Inhibitory effect of sodium pheophorbide a on carrot gray mold

    • 摘要: 脱镁叶绿酸钠 (sodium pheophorbide a) 是一种极具研发前景的光敏抑菌活性物质,为了研究将其用于防治胡萝卜灰霉病的可能性,采用菌丝生长速率法,测定了脱镁叶绿酸钠对灰葡萄孢Botrytis cinerea 的光敏抑制作用,并研究了不同质量浓度脱镁叶绿酸钠对胡萝卜灰霉病病斑扩展的影响及对胡萝卜组织可溶性蛋白含量及超氧化物歧化酶 (SOD)、过氧化物酶 (POD)、过氧化氢酶 (CAT)、苯丙氨酸解氨酶 (PAL) 活性的影响。结果表明:脱镁叶绿酸钠质量浓度以及有无光照对脱镁叶绿酸钠的抑菌活性均有显著影响 (P < 0.05),且随着脱镁叶绿酸钠质量浓度的增加,其抑制作用逐渐增强;20 mg/mL的脱镁叶绿酸钠对灰葡萄孢菌丝生长的抑制率在光照条件 (4000 lx) 下为89.19%,在黑暗条件下为72.20%;此外,20 mg/mL的脱镁叶绿酸钠处理能够有效抑制胡萝卜上灰霉病病斑的扩散,在接种后84 h内,病斑直径均显著小于其他浓度处理组 (P < 0.05),同时可以有效地保持胡萝卜组织内可溶性蛋白含量及SOD、POD、CAT和PAL 4种酶活力的稳定。综上所述,脱镁叶绿酸钠能够有效地抑制灰葡萄孢的菌丝生长,对胡萝卜灰霉病有良好的防治作用。

       

      Abstract: Sodium pheophorbide a (SPA) is a photoactivated antimicrobial active substance with great development prospects. In order to study its potential application in the control of carrot gray mold, the photoactivated antifungal effects of sodium pheophorbide a on Botrytis cinerea were determined by the mycelial growth rate method. Also, the effects of different mass concentrations of sodium pheophorbide a on the extension of carrot gray mold lesions and soluble protein content were determined. And the enzyme activities of superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), phenylalanine ammonia lyase (PAL) in carrot tissues were also analyzed. Results showed that the mass concentration and light had significant effects on the antifungal activity of sodium pheophorbide a (P<0.05). And with the mass concentration of sodium pheophorbide a increased, its inhibitory effect gradually increased. The inhibitory rate of 20 mg/mL SPA solution on the mycelium growth was 89.19% under light conditions (4000 lx) and 72.20% under dark conditions. In addition, treatment with 20 mg/mL sodium pheophorbide a solution could effectively inhibit the spread of gray mold lesions on carrots. And within 84 h after inoculation, the lesion diameter was significantly smaller than that of other groups treated with different concentrations (P<0.05). Meanwhile, it can effectively maintain the stability of the protein content, and the activities of SOD, POD, CAT and PAL in carrot. In summary, sodium pheophorbide a could effectively inhibit the mycelial growth of B. cinerea and had a good control effect on carrot gray mold.
