    线粒体呼吸链复合物III抑制剂作用机制和抗性分子机制研究进展

    Research progress on mode action and molecular resistance mechanism of complex inhibitors in cellular respiration chain

    • 摘要: 复合物Ⅲ是真核生物线粒体电子传递链上的重要功能复合体,以复合物Ⅲ为靶标开发的呼吸抑制剂被广泛应用于真菌和卵菌病害防治。根据该类杀菌剂作用位点的不同,将复合物Ⅲ抑制剂分为QoI、QiI和QioI 3类,其中QoI类杀菌剂种类较多,且抗药性问题突出。本文总结了复合物Ⅲ抑制剂的作用机制和抗性分子机制,主要包括药剂靶标蛋白细胞色素b (cyt b)G143A和F129L的氨基酸取代、旁路氧化途径中交替氧化酶基因 (AOX) 的过量表达以及运输体机制等,旨在为该类杀菌剂的合理使用提供参考。

       

      Abstract: Complex Ⅲ is an essential component of cellular respiration chain, and complex Ⅲ inhibitors are widely used for the control of fungal and oomycete diseases worldwide. According to the binding sites of the fungicides on the complex Ⅲ, they could be divided into three types, including QoI, QiI and QioI fungicides. Among them, the QoIs are the most common fungicides, which also suffers from heavy problem of fungicide resistance. This review summarizes the mode of actions and molecular resistance mechanisms of complex Ⅲ inhibitors, such as the G143A and F129L amino acid substitutions in the target protein cytochrome b (cyt b), the overexpression of alternative oxidase gene (AOX) in the alternative oxidation pathway, and the transport mechanism, aiming to provide a reference for the rational use of complex Ⅲ inhibitors.
