    草莓炭疽病菌对吡唑醚菌酯的敏感性及旁路氧化在胶孢炭疽菌对QoIs敏感性中的作用

    Characterization of sensitivity of Colletotrichum gloeosporioides from strawberry to pyraclostrobin and involvement of alternative respiration in the regulation of sensitivity

    • 摘要: 采用菌丝生长速率法测定了草莓炭疽病菌对吡唑醚菌酯的敏感性,并分析了旁路氧化与胶孢炭疽菌对QoIs类杀菌剂敏感性的关系。结果表明:无旁路氧化酶(alternative oxidase,AOX)专化性抑制剂水杨肟酸(SHAM )作用时,吡唑醚菌酯对采自浙江临安、建德、诸暨3地的65株草莓炭疽病菌菌丝生长的EC50 (EC50Q )值在0.09~24.26 μg/mL之间,平均为6.65 μg/mL;当有50 μg/mL 的SHAM存在时,其EC50EC50 (Q+S)值在0.06~48.98 μg/mL之间,平均为10.58 μg/mL。旁路氧化的相对贡献值F F=EC50Q/ EC50 (Q+S)最低为0.02,最高为11.17,平均为0.96,其中78.46%的菌株的F值AOX基因表达的时间动态。结果显示:吡唑醚菌酯处理能诱导AOX基因的表达,在处理后6~12 h内,各菌株AOX基因的表达水平均显著增强,但表达量的差异与菌株对吡唑醚菌酯的敏感性之间无显著相关性。本研究表明,旁路氧化在不同病原菌中的情况是非常复杂的。

       

      Abstract: The sensitivity of Colletotrichum gloeosporioides from strawberry to pyraclostrobin, a Qo inhibitor (QoI) fungicide, was assessed through the mycelial growth rate method and the relationship between alternative respiration and sensitivity to pyraclostrobin during mycelial growth in C.gloeosporioides was analyzed. The results showed that the medium effective concentrations (EC50Q) of 65 C.gloeosporioides isolates collected from strawberry fields in Lin'an, Jiandei and Zhuji of Zhejiang province ranged from 0.09 to 24.26 μg/mL, with an average value of 6.65 μg/mL in the absence of SHAM, a specific inhibitor of alternative oxidase. In the presence of 50 μg/mL SHAM, the EC50 (Q+S) ranged from 0.06 to 48.98 μg/mL, with an average value of 10.58 μg/mL. The factor value F=EC50Q / EC50 (Q+S) for the impact of alternative respiration on the sensitivity of isolates to pyraclostrobin ranged from 0.02 to 11.17, with an average value of 0.96; and the F value for 78.46% isolates was less than 1. The transcript levels of alternative oxidase encoding gene, AOX, in mycelia of strawberry and grape C.gloeosporioides isolates with different sensitivity to pyraclostrobin were studied through real-time quantitative PCR. The results indicated that pyraclostrobin significantly increased the AOX transcript levels after treatment for 6 to 12 h in both strawberry and grape isolates. However, no significant correlation between the AOX transcript levels and the sensitivity to pyraclostrobin was observed. The results of present study suggested that the alternative respiration pathway in different fungal pathogens is complicated.
