嗜线虫致病杆菌 SN313 次生代谢产物分离鉴定及其对南方根结线虫的生物活性

    Separation and identification of secondary metabolites from Xenorhabdus nematophila SN313 and the biological activity against Meloidogyne incognita

    • 摘要: 本研究分离得到了4个嗜线虫致病杆菌(Xenorhabdus nematophila) SN313的次生代谢产物,并测定了其杀线虫活性。通过硅胶柱层析、凝胶柱层析、高效液相色谱等技术对嗜线虫致病杆菌SN313次生代谢产物进行分离纯化,进一步采用24孔板法测定了次生代谢产物对J2时期南方根结线虫 (Meloidogyne incognita) 的毒杀活性。结果表明:共分离纯化得到4个化合物,经高分辨质谱 (HRMS)、核磁共振氢谱 (1H NMR)、碳谱 (13C NMR) 综合解析和相关文献数据比对,结合 X-射线单晶衍射法确定了其立体结构,其中化合物1是一种全新的含硝基酰胺类化合物,命名为致病杆菌酰胺 (xenornemamide);化合物2~4为已知的色胺类化合物。毒杀活性测定结果表明,化合物1、3、4对南方根结线虫的活性较好,LC50值分别为31.62、36.89、39.05 µg/mL,具备进一步研究的潜力。

       

      Abstract: This study isolated four secondary metabolites from Xenorhabdus nematophila SN313 and evaluated their nematicidal activity. Secondary metabolites were isolated and purified from X. nematophila SN313 using techniques including silica gel column chromatography, gel column chromatography, and HPLC. Their toxicity against second-stage juveniles (J2) of the Meloidogyne incognita was subsequently determined using a 24-well plate assay. The results demonstrated the isolation and purification of four compounds. Comprehensive analysis through HRMS, 1H NMR, 13C NMR, comparison with literature data, and X-ray single-crystal diffraction established their structures. compound 1 was identified as a novel nitro-containing amide, designated xenornemamide. compounds 2−4 were determined to be known tryptamine derivatives. Nematicidal activity assays revealed that compounds 1, 3, and 4 exhibited significant activity against M. incognita, with LC50 values of 31.62, 36.89, and 39.05 µg/mL, respectively. These results indicate their potential for further investigation.
