三门峡烟株内生放线菌分离及拮抗促生菌株筛选和鉴定

    Isolation of endophytic actinomycetes from tobacco plants in Sanmenxia and screening and identification of antagonistic and plant growth promoting strains

    • 摘要: 为了从烟株内筛选出具有拮抗烟草疫霉和促进植物生长特性的内生放线菌,本研究采用组织分离法对来自三门峡地区的健康烟株进行了内生放线菌的分离、纯化,并分析了烟株不同生长时期和不同组织的菌株分离情况;通过平板对峙培养法初筛和菌丝生长速率法复筛,评估内生放线菌拮抗烟草疫霉的活性;采用Salkowski比色液显色法测定菌株产3-吲哚乙酸(IAA)能力;综合形态学、生理生化特征和分子生物学鉴定确定拮抗促生菌株的分类地位。结果表明,从4个采样点的团棵期、旺长期、采收期烟株的根、茎、叶中共分离获得250株内生放线菌;烟株不同部位分离的菌株占比为:根部>叶部>茎部,分别为68.80%、19.20%和12.00%;烟株不同生长时期分离的菌株占比为:团棵期>旺长期>采收期,分别为47.20%、42.40%和10.40%;初筛获得对烟草疫霉拮抗活性60%以上的菌株共64株,复筛获得对烟草疫霉有拮抗活性的菌株8株,抑制率为16.06%~70.89%;产IAA能力的菌株7株,产量为17.15~40.09 µg/mL;结合初筛和复筛抑菌率以及IAA产量,将T-66和C-77确定为候选菌株,分别鉴定为草原链霉菌(Streptomyces pratensis)和娄彻链霉菌(S. rochei)。本研究筛选到的2个菌株为烟草黑胫病的生物防治提供了新的菌种资源。

       

      Abstract: To screen for endophytic actinomycetes with antagonistic properties against Phytophthora nicotiana and growth-promoting characteristics, this study employed tissue separation methods to isolate and purify endophytic actinomycetes from healthy tobacco plants sourced from the Sanmenxia region. Additionally, the isolation frequencies of these strains were analyzed across different plant tissues and growth stages. Initial screening was conducted using confrontation culture methods, followed by rescreening via mycelial growth rate assays to evaluate the antagonistic activity of the endophytic actinomycetes against P. nicotiana. The ability of the strains to produce 3-indoleacetic acid (IAA) was determined using the Salkowski colorimetric assay. The taxonomic status of the antagonistic and growth-promoting strains was determined through morphological, physiological, biochemical, and molecular biological characterization. A total of 250 endophytic actinomycete strains were isolated. The isolation frequencies across different plant parts were as follows: root > leaf > stem, at 68.80%, 19.20%, and 12.00%, respectively. Across different growth stages, the frequencies were: rosette growth stage > prosperous growth stage > maturation stage, at 47.20%, 42.40%, and 10.40%, respectively. Initial screening identified 64 strains with antagonistic activity exceeding 60.00% against P. nicotiana, and rescreening yielded 8 strains with inhibition rates ranging from 16.06% to 70.89%. Seven strains were found to produce IAA, with yields ranging from 17.15 to 40.09 μg/mL. Based on the combined results of initial and rescreening inhibition rates and IAA production, strains T-66 and C-77 were selected as candidate strains and were identified as Streptomyces pratensis and S. rochei, respectively. These two strains identified in this study provide new microbial resources for the biological control of tobacco black shank disease.
