HPLC-MS/MS法测定棉籽壳中11种植物生长调节剂残留

    Determination of 11 plant growth regulators in cottonseed hulls by HPLC-MS/MS

    • 摘要: 本研究通过精准筛选质谱监测离子,优化质谱参数、色谱分离条件及样品前处理方法,建立了基于高效液相色谱-三重四极杆质谱(HPLC-MS/MS)的棉籽壳中烯效唑、矮壮素、甲哌鎓、噻苯隆、敌草隆、胺鲜酯、氟节胺、吡草醚、2-硝基苯酚钠、4-硝基苯酚钠和5-硝基邻甲氧基苯酚钠共11种植物生长调节剂残留同时测定的方法,实现了不同极性目标物的有效分离与定量分析。棉籽壳用水饱和,经乙腈提取,PSA、MgSO4分散固相萃取吸附净化,多反应监测模式(MRM)分析,基质标准曲线外标法定量。结果表明:11种植物生长调节剂的检出限为0.0003~0.04 mg/kg,定量限为0.0001~0.1 mg/kg,回收率在70%~103%之间,相对标准偏差在0.30%~13%之间。利用所建立的方法分析市售棉籽壳样品,发现13份样品中共检出5种植物生长调节剂。该方法简便、灵敏度高、稳定性好,棉籽壳中噻苯隆、矮壮素、敌草隆和甲哌鎓残留情况较为突出,且普遍存在多残留现象。

       

      Abstract: In this study, a multi-residue analytical method for determination of 11 plant growth regulators (PGRs), including uniconazole, chlormequat, mepiquat, thidiazuron, diuron, diethyl aminoethylhexanoate, flumetralin, pyraflufen-ethyl, sodium 2-nitrophenoxide, sodium 4-nitrophenoxide and sodium 5-nitroguaiacolate, in cottonseed hulls was developed based on high-performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS). After systematic optimization of sample preparation and instrumental parameters, the method achieved effective separation and quantitative analysis of the target compounds with different polarities. The cottonseed hull samples were hydrated with water, extracted with acetonitrile, cleaned-up by dispersive solid-phase extraction (d-SPE) with PSA and MgSO4, then determined with HPLC-MS/MS at multiple reaction monitoring (MRM) mode, and quantified by the external standard method with matrix-matched calibration curves. The results demonstrated that the limits of detection (LODs) of 11 PGRs ranged from 0.0003 to 0.04 mg/kg and the limits of quantification (LOQs) ranged from 0.0001 to 0.1 mg/kg. The average recoveries varied from 70% to 103%, and the relative standard deviations (RSDs) were 0.30% to 13%. The method is simple and has high precision and robust stability. It was applied to analyze market samples, and the residues of 5 PGRs were detected in 13 samples. Notably, chlormequat chloride, mepiquat chloride, thidiazuron, and diuron exhibited a relatively high detection frequency, inferring that the multiple residues of PGRs are common in cottonseed hulls.
