沸石咪唑酯骨架负载嘧霉胺载药体系的制备及其对番茄灰霉病的防治效果

    Preparation of ZIF-8 loaded pyrimethanil delivery system and its control efficiency against tomato gray mold

    • 摘要: 嘧霉胺作为一种苯氨基嘧啶类杀菌剂,对番茄灰霉病具有良好的防治效果,但其传统剂型存在利用率低、易造成环境污染等问题。为提高嘧霉胺对番茄灰霉病的防治效果并实现其智能化释放,构建了一种能响应病害酸性微环境的金属-有机框架载药系统。采用一锅法成功合成了基于沸石咪唑酯骨架(ZIF-8)的形貌规整、稳定性良好的嘧霉胺负载纳米颗粒(Pyr@ZIF-8),载药量为14.72%,平均粒径为(346.80 ± 5.44) nm。该载药系统表现出显著的pH值依赖性释放动力学特征,在pH 5和pH 9条件下的累积释放率分别为84.94%和53.60%,这种酸快碱慢的pH响应特性主要受骨架溶蚀机制驱动。平板抑制试验结果显示,Pyr@ZIF-8的EC50值(6.68 mg/L)低于嘧霉胺(7.93 mg/L)、ZIF-8载体(9.66 mg/L)和Pyr + ZIF-8物理混合组(7.40 mg/L)。盆栽防效试验结果表明,药后14 d,Pyr@ZIF-8的防治效果分别为保护作用77.00%和治疗作用65.27%,显著高于商品化制剂40%嘧霉胺悬浮剂(分别为36.41%和24.26%)。Pyr@ZIF-8成功实现了嘧霉胺的pH响应型控释,并通过纳米载体增效作用显著提高了其生物活性,为实现农药的精准高效施用提供了新途径。

       

      Abstract: Pyrimethanil, a phenylamino-pyrimidine fungicide, exhibits effective control against tomato gray mold (Botrytis cinerea), but its conventional formulations face challenges such as low utilization efficiency and potential environmental pollution. To improve control efficacy of pyrimethanil against tomato gray mold and achieve intelligent release, this study developed a metal-organic framework (MOF) carrier system responsive to the acidic microenvironment typically induced by fungal infections. The pyrimethanil-loaded nanoparticles (Pyr@ZIF-8) with regular morphology and good stability were successfully synthesized via a one-pot method based on zeolitic imidazolate framework-8 (ZIF-8), with a drug loading capacity of 14.72% and an average particle size of (346.80 ± 5.44) nm.The drug delivery system demonstrated significant pH-dependent release kinetics, with cumulative release rates of 84.94% and 53.60% at pH 5 and pH 9, respectively. This pH-responsive behavior, characterized by faster release under acidic conditions and slower release under alkaline conditions, was primarily driven by the framework erosion mechanism. Plate inhibition assays revealed that Pyr@ZIF-8 exhibited the lowest EC50 value (6.68 mg/L) compared to free pyrimethanil (7.93 mg/L), the ZIF-8 carrier (9.66 mg/L), and the physical mixture of Pyr + ZIF-8 (7.40 mg/L). The pot experiment results showed that 14 days after application, the control efficacy of Pyr@ZIF-8 was 77.00% (protective effect) and 65.27% (curative effect), significantly higher than that of the commercial 40% pyrimethanil suspension agent (36.41% and 24.26%, respectively).This pH-responsive controlled-release system significantly enhances the biological activity of pyrimethanil through nano-carrier synergistic effects, providing a promising strategy for the precise and efficient application of pesticides.
