    棉铃虫谷胱甘肽S-转移酶的活性分布和发育期变化及植物次生物质的诱导作用

    Glutathione S-Transferases(GSTs) in Helicoverpa Armigera : Subcellular and Tissue Distribution of Activity, Developmental Changes and Induction of Allelochemicals

    • 摘要: 对棉铃虫 Helicoverpa armigera (Hübner)谷胱甘肽S-转移酶(GSTs)发育期活性进 行了跟踪测定。从卵期开始,GSTs活性呈上升趋势,6龄幼虫达到最高,然后逐渐下降,但化蛹初期仍维持较高水平;用培养基混药法研究发现,芸香苷和2-十三烷酮诱导种群5龄时 活性即达到峰值。6龄幼虫脂肪体GSTs活性最高,中肠次之,头部和表皮最低。用1-氯-2, 4-二硝基苯(CDNB)作底物时,中肠GSTs的 Km值最小。5龄幼虫中肠匀浆液中,GSTs活性90%以上集中在细胞液层,在微粒体、线粒体和细胞碎片层的分布均不超过5%;芸香苷诱 导种群3龄幼虫微粒体层GSTs活性分布明显比对照种群减少,而上清液层则比对照种群 有所增高。

       

      Abstract: The Activity of GSTs in different tissues, developmental stages, subcellular fra ctions and induction by allelochemicals was determined in cotton bollworm, Helicoverpa armigera (Hübner). GSTs activity toward 1-chloro-2,4-dinitrobenze ne ( CDNB) was detected in the egg. It increased throughout the larval stages and up to the highest in the sixth instar larvae. The activity of GSTs from the larvae fed on artificial diet containing 50 mg/kg rutin or 2-tridecanone reach ed t he peak at the fifth instar. Enzyme activity was found in fat body, midgut, head and cuticle of the sixth instar larvae. The GSTs activity in fat body was the h ighest in above tissues. The Km values for CDNB conjugation varied with diff erent tissues, and the K m values of GSTs in the midgut was the lowest (8.5 mmol/L). The supernatant fraction possessed more than 90% of total activities of GSTs in the homogenate of midguts of the fifth instar larvae. The GSTs activity in microsome fraction from the third instar larvae induced by rutin was obviousl y lower than that fed only on artificial diet, but the enzyme activity in su pernatant fraction increased.
