    氯嘧磺隆抗体制备的研究

    Preparation of Polyclonal Antibodies to Chlorimuron-ethyl

    • 摘要: 以合成的半抗原与牛血清白蛋白(BSA)的联结物为免疫抗原,获得对氯嘧磺隆高亲合力的兔抗血清,建立了氯嘧磺隆的直接竞争酶联免疫检测方法。抗血清效价为6.4×104,IC50值为10.9 ng/mL,测定的线性范围是0.4 ~295.4 ng/mL。在河水中的添加回收率为87.9% ~115.7%。结构相似的常用磺酰脲类除草剂与氯嘧磺隆抗体的交叉反应率为0.1% ~4.8%。所建立的方法可用于河水中氯嘧磺隆的残留检测。

       

      Abstract: A direct competitive enzyme immunoassay for the quantitative analysis of chlorimuron-ethyl was developed. Polyclonal antibodies(PAbs) were prepared from rabbits immunized with hapten- protein conjugates. The titer of antiserum was 6.4×104,IC50 was 10.9 ng/mL. Chlorimuron-ethyl could be quantitated in the range of 0.4 ~295.4 ng/mL.Average recovery of chlorimuron-ethyl in river water was 87.9% ~115.7%.The cross-reactivity of PAbs of chlorimuron-ethyl with other analogues were 0.1% ~4.8%. This assay can be applied for the quantitation of chlorimuron-ethyl residue in river water.
