    The Separation,Purification and Bioassay of the Effective Metabolites of SPRI-231

    • 摘要: 从广西南宁地区采集的土壤里筛选出一株能够产生抗菌活性物质的链霉菌株(streptomyces), 菌号为SPRI-231。通过溶剂浸提、大孔树脂吸附、柱层析、薄层层析、高效液相色谱等方法分离得到两个具有抑菌活性的化合物,分别命名为SPRI-231(Ⅰ)和SPRI-231(Ⅱ),经过UV、IR、ESI-MS、ESI-HRMS、元素分析等鉴定方法,判定其分子式分别为C51H91N3O21和C52H93N3O21。活体和离体生物活性测定发现其具有较好的抗菌效果,500 mg/L SPRI-231(Ⅱ)精提物对黄瓜灰霉病菌和黄瓜炭疽病菌的抑制率分别为83.02%和90.40%。

       

      Abstract: For the screening of new agro-antibiotics, a strain of streptomycetes was isolated from the soil sample collected in Guangxi Province and named SPRI-231. The strain's active metabolites of the strain were isolated and purified with solvent extraction, silica gel chromatography, TLC, HPLC etc. The pure components of compound SPRI-231 (Ⅰ) and SPRI-231 (Ⅱ) were obtained. Based on physico-chemical properties, as well as UV、IR、ESI-MS、ESI-HRMS and elemental analysis, their molecular formula were confirmed as C51 H91 N3 O21 and C52 H93 N3 O21 ,respectively. These two compounds have good effect in protecting plant fungi diseases, the inhibition rate of refined extracts of SPRI-231(Ⅱ) at 500 mg/L against cucumber Botrytis cinerea and cucumber Colletotrichum lagenarium were 83.02% and 90.40%,respectively.
