    氯氟氰菊酯及其代谢物的人工抗原合成与多克隆抗体的制备

    Synthesis of Artificial Antigen and Preparation ofPolyclonal Antibodies against Cyhalothrin and it's Metabolites

    • 摘要: 于氟氰菊酯分子的酸部分连接4-氨基丁酸间隔臂(HCA1)、醇部分连接丁二酸酐间隔臂(HCB2),分别合成了两种不同的半抗原,通过碳二亚胺法将HCA1和HCB2分别与牛血清蛋白(BSA)偶联制备了人工抗原HCA1-BSA和HCB2-BSA,通过混合酸酐法将HCA1、HCAS 和HCB2分别与卵清蛋白(OVA)偶联制备包被抗原,两种人工抗原通过免疫新西兰大白兔制备相应的多克隆抗体。结果表明,用HCA1-BSA获得的多克隆抗体对氯氟氰菊酯和氯氟氰菊酸有免疫反应性(IC50值分别为33.12 和0.95 mg/L),用HCB2-BSA获得的多克隆抗体对氯氟氰菊酯几乎没有免疫反应性。

       

      Abstract: Two types of haptens,the carboxylated propyl amino derivative of the acid moiety (HCA1) that is the hydrolyzed product of cyhalothrin,and the carboxylated ethyl carbonyl derivative of the alcohol moiety (HCB2) were synthesized.HCA1 and HCB2 were conjugated with the carrier proteins bovine serum albumin (BSA) by the active ester method to obtain the artificial immunogens.HCA1、HCAS and HCB2 were conjugated with ovalbumin (OVA) by the mixed anhydride method to obtain the coating antigens respectively.The polyclonal antibodies (PAbs) were obtained after the immunogens immunized to the New Zealand rabbits.One PAbs,which raised against the former hapten HCA1,was reactive with cyhalothrin and cyhalothric acid (IC50:33.12 mg/L,0.95 mg/L).However,the other PAbs,which raised against the later hapten HCB2,was not particularly reactive with cyhalothrin.
