    Study on the Application of Multifactor ExperimentalDesigns in Pesticide Microemulation Development

    • 摘要: 以2%阿维菌素微乳剂为研究对象,对正交设计与均匀设计两种多因素试验设计方法在农药微乳剂配方研制中的应用可行性进行了研究。采用固定表面活性剂总用量,保留一种表面活性剂不作为自变量,其余表面活性剂均作为自变量,同时将助表面活性剂、防冻剂作为自变量的方法设计试验方案;因变量选择微乳剂透明温度区域(TTS)的下端温度(TL)、上端温度(TH)及整个区域范围(TR=TH-TL)。以TH作为因变量,对正交设计测试数据进行方差分析与极差分析,所获得优化配方的TTS为-15.0℃~66.5℃;对均匀设计测试数据采用偏最小二乘法,同时对TH及TR进行回归,模型决定系数R2分别为0.994 5和0.985 1,TH、TR预测结果分别为71.5℃和 87.1℃,实测结果分别为71.6℃和87.3℃;Turbiscan 扫描结果表明,应用正交设计与均匀设计优化得到的制剂在54℃下热贮14 d,体系均相当稳定。研究结果表明:(1)正交设计与均匀设计均可成功应用于农药微乳剂配方研制,可大幅度减轻工作量,快速获得性能优异的制剂配方;(2)将微乳剂TTS作为正交设计与均匀设计试验时的考察指标是合适的,TTS可拆分成TL、TH及TR 3个因变量。

       

      Abstract: The application of two multifactor experimental designs,orthogonal design (OD) and uniform design (UD),for developing pesticide microemulsion (ME) were investigated through a case study of abamectin 20 ME.In experimental design,the total dosage of surfactants was fixed,each of the used surfactant was as independent variable except one,and co-surfactant and antifreeze were also as independent variables.Transparency temperature section (TTS) of ME was tested as response in OD and UD experiments,and its low point temperature (TL),high point temperature (TH) and range (TR=TH-TL) were selected as dependent variables.Analysis of variance and rang R were introduced to processing OD experiment data,and TH was selected as dependent variable; TTS of abamectin 20 ME optimized by OD was -15.0℃~66.5℃.Partial Least Squares Regression was introduced to processing UD experiment data,and TH and TR were selected as dependent variables and optimized together during modeling; two regression equation models were obtained,and R2 of models were 0.9945,0.9851,respectively; the prediction values of TH and TR were 71.5℃ and 87.1℃,respectively,while actual tested values were 71.6℃ and 87.3℃,respectively.Turbiscan scanning showed that the preparations optimized by OD and UD were very stable during stored 14 d successively at 54℃.Results showed that: (1) OD and UD could be successfully applied to pesticide ME development.The two experimental designs could substantially lighten formulators' workload,and help formulators to quickly obtain recipe with good performance.(2) TTS was a appropriate response variable for OD and UD,and it could be split into three dependent variables: TL,TH and TR.
