    氯虫苯甲酰胺对小菜蛾鱼尼丁受体基因mRNA表达量的影响

    Effects of chlorantraniliprole on expression abundance of ryanodine receptor gene mRNA from Plutella xylostella

    • 摘要: 采用实时荧光定量PCR研究比较了氯虫苯甲酰胺、甲氨基阿维菌素苯甲酸盐和溴虫腈对小菜蛾鱼尼丁受体基因mRNA表达量的影响,研究分析了低剂量氯虫苯甲酰胺连续处理和间断处理对小菜蛾鱼尼丁受体基因表达量的影响,以及对氯虫苯甲酰胺产生抗性的小菜蛾与敏感小菜蛾鱼尼丁受体基因表达量的差异。结果表明:氯虫苯甲酰胺可提高小菜蛾鱼尼丁受体基因表达量,而甲氨基阿维菌素苯甲酸盐和溴虫腈对其表达量均无影响;0.05和0.01 mg/L的氯虫苯甲酰胺均可使小菜蛾鱼尼丁受体基因表达量增加;对氯虫苯甲酰胺产生抗性的小菜蛾鱼尼丁受体基因表达量是敏感小菜蛾的5.79倍。研究结果有助于阐明氯虫苯甲酰胺的作用机理,并可能为筛选作用于鱼尼丁受体的新化合物提供参考。

       

      Abstract: The expression abundance of ryanodine receptor(RyR)mRNA from Putella xylostella(L.)larvae treated with chlorantraniliprole,emamectin benzoate and chlorfenapyr respectively were studied by quantitive RT-PCR method.The results showed that chlorantraniliprole could induce the expression of RyR gene from P.xylostella,and emamectin benzoate and chlorfenapyr had no effects on the expression of RyR gene . In order to further identify the induction of chlorantraniliprole to RyR gene,the relative expression abundance of RyR gene from larvae treated continuously and discontinuously with 0.01 mg/L chlorantranilipole were analyzed,and the RyR gene expression abundance from P.xylostella that resistant to chlorantraniliprole was also analyzed.The results suggested that 0.05 mg/L and 0.01 mg/L chalorantraniliprole could induce the expression level of RyR gene from P.xylostella in both treatment;Moreover,the relative expression level of RyR gene from chlorantraniliprole resistant strain was 5.79-fold comparing with that of the susceptible strain.The results would be helpful to indicate the mechanism of chlorantraniliprole,and the results would be possibly to helpful to screen compounds targeting at RyR.
